Cyclin D has been shown in many cancer types to be misregulated. Well established for their oncogenic properties, the cyclins, and the cyclin-dependent kinases (CDK's) they activate, have been the focus of major research and development efforts over the past decade. The methods by which the cyclins are misregulated are widely variable, ranging from genomic amplification to changes in promoter methylation. The 11;14 translocation is frequently associated with mantle cell lymphoma and may be found in other hematologic tumors such as multiple myeloma, plasma cell leukemia and chronic lymphocytic leukemia. This translocation results in the fusion of the immunoglobulin heavy chain gene (IGH), located at chromosome region 14q32, with the cyclin D1 (CCND1) gene, located at chromosome region 11q13.
