The ready-to-use IDBRAF-CNS kit is a multiplexed and sensitive tool for the detection and quantification by digital PCR (dPCR) of V600E mutation and the tyrosine kinase domain (TKD) duplication of the BRAF gene (B-Raf proto-oncogene, serine/threonine kinase).
TKD duplication of the BRAF gene, located at 7q34, systematically accompanies its fusion with the KIAA1549 gene (7q34). The detection of the KIAA1549-BRAF fusion is, according to the WHO recommendation (1), necessary for the classification of certain brain tumors in children and young adults.
The KIAA1549-BRAF fusion is a feature of pilocytic astrocytoma, but it has also been shown in rare cases of gangliogliomas, oligodendroglial tumors co-deleted for 1p / 19q.
BRAF V600E mutations are observed in many low-grade glioma subtypes, including 20% fibrillar astrocytomas, 50% gangliogliomas, 75% pleomorphic xanthoastrocytomas and 5% pilocytic astrocytomas.
This optimized kit provides a multiplex solution allowing, for each sample, to simultaneously amplify, in one PCR reaction, the V600E mutations (c.1799T>A) and the fusion of the KIAA1549-BRAF genes (Ref: IDBRAF-CNS(b)-25, IDBRAF-CNS(s)-25).
