Invitrogen™ Amplex™ Red Phospholipase D Assay Kit

The ELF™ 97 Endogenous Phosphatase Kit can be used to detect phosphatase activity with the ELF™ 97 phosphatase substrate that upon hydrolysis produces a bright and photostable yellow-green fluorescent precipitate at the site of enzymatic activity. This fluorescent precipitate has several unique spectral characteristics, including an extremely large Stokes shift, 180 nm that makes it easily distinguishable from endogenous fluorescence.

PLD activity is monitored indirectly using 10-acetyl-3,7-dihydroxyphenoxazine (Amplex™ Red reagent), a sensitive fluorogenic probe for hydrogen peroxide. PLD converts the phosphatidylcholine (lecithin) to choline, which is then oxidized by choline oxidase to betaine and hydrogen peroxide. In the presence of horseradish peroxidase, huydrogen peroxide reacts with the Amplex™ Red reagent in a 1:1 stoichiometric ratio to generate the highly fluorescent product resorufin.
