The ready-to-use IDPIK3CA kit is a multiplexed and sensitive tool for the detection and quantification by digital PCR (dPCR) of mutations in the PIK3CA gene (phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha).
The PI3K/AKT pathway plays a key role in several cellular functions, including apoptosis and proliferation. This pathway can be directly activated by activation of EGFR or via the RAS proteins. Colorectal, cerebral and gastric cancers exhibit a high rate of PIK3CA gene mutations with respective frequencies of 32%, 27% and 25%. Breast and lung cancers have a relatively lower rate of PIK3CA mutations (8% and 4% respectively). These somatic mutations are dispersed in most exons but are mainly found in the kinase and helical domains of the PIK3CA subunits, located on exon 9 (E542K, E545K, Q546K) and 20 (H1047R, H1047L).
This optimized kit provides a multiplex solution allowing, for each sample, to simultaneously amplify, in one PCR reaction, the PIK3CA‘s mutations at E542K, E545K, Q546K, H1047R and H1047L (Ref: IDPIK3CA(s)-25).
